Light-Sheet Microscopy with Bessel beams and STED

 
Background and Problem: The extended ring system of the Bessel beam is required for self-re-construction, but excites fluores-cence inside the object above and below the focal plane and thus reduces image contrast.
Approach 1: Using femto-second lasers, fluorescence inside the strongly scattering medium is excited by a two-photon process, leading to strong suppression of the unwanted fluorescence in the ring system.
Approach 2: The principle of stimulated emission by depletion (STED) is applied for the first time to self-reconstructing beams. The un-wanted fluorescence in the Bessel beam’s ring system is depleted by a second self-reconstructing Bessel beam with a helical phase (charge number L = 3).
